NEW METHOD FOR CELL CYCLE SYNCHRONIZATION IN AMOEBA PROTEUS CULTURE

A method to synchronize cell cycle in the culture of Amoeba proteus is proposed. Amoeba cells were kept in quail egg albumen and thus having been fed by the pinocytosis, then thoroughly washed from the protein and examined at stained slides in different periods of time after washing. It appeared that practically all the cells on the slide corresponding to certain time after feeding/washing underwent the same stage of cell cycle — either interphase or one of the phases of mitosis depending on time. The method of amoebae feeding by the pinocytosis also allows avoiding negative effect of food remains in the cell cytoplasm, thus providing the higher quality of stained slides.

HEAT SHOCK PROTEINS OF FREE-LIVING CILIATES AND THEIR IMPACT ON CELL ADAPTATION TO SALINITY STRESS

The paper describes the comparative analysis of our own experimental data concerning the chaperone activity of 70 kDa heat shock protein (HSP70) in three free-living ciliate species, Paramecium jenningsi, Tetrahymena pyriformis, Paramecium nephridiatum, belonging to the eufreshwater, metafreshwater and euryhaline ecological groups of unicellular organisms. These ciliates revealed different protecting strategies and adaptive potential of their chaperon defensive system regarding to the salinity stress. We infer that the constitutive level of HSP70 content in free-living ciliates reflects the environmental conditions of locations, where from these species originated. Consequently, when evaluating the ecological importance of constitutive HSP70 level in hydrobionts, it is necessary to take into account not only contemporary conditions of their existence, but the chaperone system “memory” as well.

Prosicuophora cyclostomatus n. sp. and Nyctotheroides bambuiensis n. sp. are two new ciliate species isolated from the rectums of Bufo regularis collected in the vicinity of Bambili and Bambui. The morphological characteristics of these Protozoa are described and their relation to similar species is discussed.

The article’s subject is the life story of Prof. Butschli and his main achievements in Protozoology and Cytology as well as his relationship with Russian scientists. For the first time the unknown history of his grave and the grave monument are revealed. It has been found out that the grave is located in Karlsruhe but not in Heidelberg, as thought earlier, where the scientist spent 42 most productive years of his life. The article is illustrated with several unique photos.
